Although both the answer and definition are singular nouns, I cannot understand how one could define the other.
'uncle has to look out for' is the wordplay.
'uncle' becomes 'sam' (Uncle Sam is an example).
'has' says to put letters next to each other.
'to look' becomes 'see'.
'out' means to remove the last letter.
'for' becomes 'e' (I can't explain this - if you can you should believe this answer much more).
'see' with its last letter taken off is 'se'.
'sam' after 'se' is 'sesam'.
'sesam'+'e'='SESAME'
