I believe the answer is:

vacuum

I'm a little stuck... Click here to teach me more about this clue!

'emptiness' is the definition.
(vacuum is a kind of emptiness)

'holiday for two of you at beginning of may' is the wordplay.
'holiday for' becomes 'vac' (I can't explain this - if you can you should give a lot more credence to this answer).
'two of' says to double the letters (I've seen 'two' mean this).
'you' becomes 'u' (I've seen this in other clues).
'at' means one lot of letters go next to another.
'beginning of' says to take the initial letters.
The first letter of 'may' is 'm'.
'u' duplicated is 'uu'.
'vac'+'uu'+'m'='VACUUM'

'of' acts as a link.

(Other definitions for vacuum that I've seen before include "Nature abhors one" , "Empty space - nature abhors it" , "cleaner" , "Space containing no matter" , "Area containing no matter" .)

I've seen this clue in The Hindu.
