Although both the answer and definition are singular nouns, I can't understand how one could define the other.

'run-through for a seamstress?' is the wordplay.
'run-through' becomes 'rehearsal' (I've seen this before).
'for' says to put letters next to each other (I've seen this in other clues).
'a seamstress?' becomes 'dress' (I am not sure about this - if you are sure you should believe this answer much more).
'rehearsal' after 'dress' is 'DRESS REHEARSAL'.
