Although both the answer and definition are singular nouns, I can't understand how one could define the other.

'party note resistance of' is the wordplay.
'party' becomes 'labour' (Labour Party is an example).
'note' becomes 'e' (musical note).
'resistance of' becomes 'r' (I am not sure about this - if you are sure you should believe this answer much more).
'labour'+'e'+'r'='LABOURER'
