Although both the answer and definition are plural nouns, I can't understand how they can define each other.

'tricolour's first among equals' is the wordplay.
'tricolour's first' becomes 't' (1st letter of 'tricolour').
'among' means one lot of letters goes inside another (inserted letters are placed among others).
'equals' becomes 'peers' (one's peers are one's equals).
't' inserted inside 'peers' is 'PETERS'.
