antagonise

'needle' is the definition.
(both can mean to provoke or irritate somebody)

'turning one against' is the wordplay.
'turning' indicates anagramming the letters.
'one'+'against'='oneagainst'
'oneagainst' anagrammed gives 'ANTAGONISE'.

(Other definitions for antagonise that I've seen before include "Get on the wrong side of" , "anger" , "Cause opposition" , "Provoke hostility" , "Bug" .)
