Although both the answer and definition are singular nouns, I can't see how one could define the other.

'leading lady in' is the wordplay.
'leading' becomes 'ur' (I can't justify this - if you can you should believe this answer much more).
'lady' becomes 'she' ('she' refers to a woman).
'in' means one lot of letters goes inside another.
'ur' enclosing 'she' is 'USHER'.

'the' acts as a link.
