Although both the answer and definition are singular nouns, I don't understand how one could define the other.

'help with a test?' is the wordplay.
'help' becomes 'el' (I can't justify this - if you can you should give a lot more credence to this answer).
'with' says to put letters next to each other.
'a test?' becomes 'MOT' (as in an MOT test for a vehicle).
'el' after 'mot' is 'MOTEL'.

'of' acts as a link.
