Although both the answer and definition are singular nouns, I can't see how they can define each other.

'fun-loving youth' is the wordplay.
'fun' becomes 'play' (synonyms).
'loving youth' becomes 'boy' (male youth. I am not sure about the 'loving' bit.).
'play'+'boy'='PLAYBOY'

'in' acts as a link.
