Although both the answer and definition are singular nouns, I don't see how one could define the other.

'imprison drug dealer' is the wordplay.
'imprison' becomes 'pen' (to pen in is to imprison).
'drug dealer' becomes 'pusher' (someone who sells or pushes drugs).
'pen'+'pusher'='PENPUSHER'
